产品简介：一种广谱抗生素,能有效抑制RNA聚合酶和阻断大肠杆菌RNA合成。Rifampicin is an antibiotic that potently inhibits RNA polymerase and blocks RNA synthesis in Escherichia coli. The compound has been shown to inhibit α-synuclein fibrillation and disaggregate fibrils in a concentration-dependent manner. Data indicates that Rifampicin can activate the nuclear pregnane X receptor (PXR), which affects cytochrome P450, and the activity of glucuronosyltransferases and P-glycoprotein. Rifampicin has been shown to enhance CYP2C-mediated metabolism, affect compounds that are transported by P-glycoprotein and metabolized by CYP3A4, and has been used in tuberculosis studies。
溶解性：溶于水（1.3mg/mL pH 4.3， 2.5mg/mL pH 7.3)、DMSO（100mg/mL）和氯仿（50mg/mL）
储备液保存：-80°C, 6 months; -20°C, 1 month。
体内实验：1、请依序添加每种溶剂： 10% DMSO→40% PEG300→5% Tween-80→45% Saline Solubility: 2.5 mg/mL (3.04 mM); 澄清溶液; 超声助溶 此方案可获得 2.5 mg/mL 的澄清溶液。 以 1 mL 工作液为例，取 100 μL 25.0 mg/mL 的澄清 DMSO 储备液加到 400 μL PEG300 中，混合均匀；再向上述体系中加入50 μL Tween-80，混合均匀；然后再继续加入 450 μL 生理盐水 定容至 1 mL。 2、请依序添加每种溶剂： 10% DMSO→90% (20% SBE-β-CD in Saline) Solubility: ≥ 2.5 mg/mL (3.04 mM); 澄清溶液 此方案可获得 ≥ 2.5 mg/mL（饱和度未知）的澄清溶液。 以 1 mL 工作液为例，取 100 μL 25.0 mg/mL 的澄清 DMSO 储备液加到 900 μL 20% 的 SBE-β-CD 生理盐水水溶液 中，混合均匀。 ＜1mg/ml表示微溶或不溶。 普西唐提供的所有化合物浓度为内部测试所得，实际溶液度可能与公布值有所偏差，属于正常的批间细微差异现象。 请根据产品在不同溶剂中的溶解度选择合适的溶剂配制储备液；一旦配成溶液，请分装保存，避免反复冻融造成的产品失效。
靶点：RNA polymerase
体外研究：Rifampicin (100 microg/mL) can block the functional activity of P-glycoprotein. Rifampicin is not a substract for P-glycoprotein. The mechanism of rifampicin resistance is unassociated with the functional activity of P-glycoprotein.
体内研究：Rifampicin (200, 400 mg/kg) can induce fatty liver at high concentration. Rifampicin (30 mg/kg,i.p.) treatment of S464P biofilms in vivo results in a slight decline, but earlier rebinds in bioluminescence from these catheters compared with the parental signal, whereas rifampicin has no affect on bioluminescence in mice infected with mutant H481Y.
