产品描述：基本信息 产品编号： H10479 产品名称： H100 CAS: 643727-55-3   储存条件 粉末 -20℃ 四年 分子式： C18H16N2O6S 溶于液体 -80℃ 一年 分子量： 388.39     化学名：    Solubility (25°C)：   体外:   DMSO   Ethanol   Water   体内(现配现用):   ＜1mg/ml表示微溶或不溶。 普西唐提供的所有化合物浓度为内部测试所得，实际溶液度可能与公布值有所偏差，属于正常的批间细微差异现象。 请根据产品在不同溶剂中的溶解度选择合适的溶剂配制储备液；⼀旦配成溶液，请分装保存，避免反复冻融造成的产品失效。   生物活性 产品描述 一种氯离子运输(Cl- transport)抑制剂，可以部分抑制氯离子载体NaK2Cl cotransporter和Band 3 anion exchanger，但是对KCl cotransporter无作用。 靶点 Cl- transport   体外研究 H100 is a Cl- transport inhibitor, with partial effects against both the NaK2Cl cotransporter (NKCC) and the Band 3 anion exchanger (AE),but no effect against KCl cotransporter (KCC),in human erythrocytes.H100 (0.1mM) shows 63% and 74% inhibition of NKCC and AE in human erythrocytes.   推荐实验方法(仅供参考) 细胞实验:   35SO4 2- efflux is used as a convenient measure of the activity of the Band 3 anion exchanger (AE).Erythrocytes are suspended into MBS containing Na2SO4 rather than NaCl and incubated at 37℃ for 30 min.The cell suspension is then centrifuged (3,000g,5 min) and the supernatant removed.The above procedure is repeated twice to ensure the intracellular replacement of Cl- with SO4 2-.To load the erythrocytes with radiolabel,the packed cells are re-suspended to approximately 50% haematocrit in a solution containing 1 part SO4 2- MBS and 9 parts of a medium containing (in mM) 300 sucrose and 10 MOPS (pH 7.4,300±5 mOsm) and placed in a microcentrifuge tube.10μCi of 35SO4 2- is then added and the suspension incubated at 37℃ for 1h.At the end of this period,the cells are then washed four times by centrifugation (10,000g,10s) in ice-cold SO4 2- MBS.All inhibitors (H100) are dissolved in either DMSO or MBS and are added to the cell suspensions prior to the addition of the radioisotope.
