VR23 produces a synergistic effect in killing multiple myeloma cells, including those that were resistant to PS-341.VR23 as a structurally novel proteasome inhibitor with desirable properties as an anticancer agent. 体内研究 VR23 shows effective antitumor and antiangiogenic activities in mice.   推荐实验方法(仅供参考) 激酶实验： 蛋白酶体试验 在96孔聚合板上以指数生长的细胞用不同浓度的药物或空白对照处理6小时。用0.5% NP40缓冲液提取的蛋白酶体与等量样品在100μL总体积中混合，然后与25μmol/L荧光底物(特异性LRR用于测定胰蛋白酶样活性，特异性LLE用于测定半胱天冬酶活性，特异性SUVY用于测定糜蛋白酶样活性)在黑底96孔板中于37°C下培育。荧光性在360nm (激发) 和480nm (发射)波长下每5分钟监测一次。   细胞实验:   细胞系 RPMI 8226，KAS 6，184B5，和 MCF10A 细胞；耐-bortezomib的 RPMI 8226 细胞和耐-bortezomib的 ANBL6 细胞 浓度 ~20μM 方法 进行克隆和SRB试验。IC50值使用S形剂量响应曲线(可变斜率)通过Graph Pad Prism V 4.02版进行计算。   动物实验:   动物模型 移植 MDA-MB-231 或 RPMI 8226 癌细胞的 ATH490无胸腺小鼠 剂量 30mg/kg 给药处理 i.p.
