体内实验：1、请依序添加每种溶剂： 10% DMSO→40% PEG300→5% Tween-80→45% Saline Solubility: ≥ 2.5 mg/mL (7.61 mM); 澄清溶液 此方案可获得 ≥ 2.5 mg/mL（饱和度未知）的澄清溶液。 以 1 mL 工作液为例，取 100 μL 25.0 mg/mL 的澄清 DMSO 储备液加到 400 μL PEG300 中，混合均匀；再向上述体系中加入 50 μL Tween-80，混合均匀；然后再继续加入 450 μL 生理盐水 定容至 1 mL。 2、请依序添加每种溶剂： 10% DMSO→90% (20% SBE-β-CD in Saline) Solubility: ≥ 2.5 mg/mL (7.61 mM); 澄清溶液 此方案可获得 ≥ 2.5 mg/mL（饱和度未知）的澄清溶液。 以 1 mL 工作液为例，取 100 μL 25.0 mg/mL 的澄清 DMSO 储备液加到 900 μL 20% 的 SBE-β-CD 生理盐水水溶液 中，混合均匀。 3、请依序添加每种溶剂： 10% DMSO→90% Corn Oil Solubility: ≥ 2.5 mg/mL (7.61 mM); 澄清溶液 此方案可获得 ≥ 2.5 mg/mL（饱和度未知）的澄清溶液，此方案实验周期在半个月以上的动物实验酌情使用。 以 1 mL 工作液为例，取 100 μL 25.0 mg/mL 的澄清 DMSO 储备液加到 900 μL玉米油中，混合均匀。 ＜1mg/ml表示微溶或不溶。 普西唐提供的所有化合物浓度为内部测试所得，实际溶液度可能与公布值有所偏差，属于正常的批间细微差异现象。 请根据产品在不同溶剂中的溶解度选择合适的溶剂配制储备液；⼀旦配成溶液，请分装保存，避免反复冻融造成的产品失效。

体外研究：AZ32 is a next-generation blood-brain barrier (BBB)-penetrating ATM inhibitor. AZ32 blocks the DNA damage response and radiosensitized GBM cells in vitro.
体内研究：AZ32, with enhanced BBB penetration, is highly efficient in vivo as radiosensitizer in syngeneic and human, orthotopic mouse glioma model compared with AZ31. AZ32 is a specific inhibitor of the ATM kinase that possesses good BBB penetration in mouse. Following a single oral dose of AZ32 (200 mg/kg) in mice, the free-brain concentrations of AZ32 are in excess of the cellular IC50 for approximately 22 hours.
细胞实验：细胞系:U1242细胞 浓度:3 μM 处理时间:48 h 方法:用AZ32 (3 μM)处理细胞并进行辐射(2 Gy)。48小时后，收集细胞固定，进行免疫细胞化学分析。
动物实验：动物模型:原位GL261胶质瘤模型（将转染了luc-red的GL261细胞通过颅内注射到C57bl6小鼠中，构建此模型） 剂量:200 mg/kg 给药处理:p.o.
